In addition, 4-FPH has been used as an intermediate in the synthesis of other compounds, such as 4-fluoro-3-hydroxyphenylacetic acid (4-FHPAA), which has been used in the treatment of Alzheimer’s disease.

Synthesis Techniques

4-Fluoropiperidines can be synthesized efficiently through aza-Prins cyclization, a process that combines aldehydes with N-tosyl homoallylamine. This method is advantageous for its simplicity, convenience, and cost-effectiveness, yielding high cis-selectivity and good yields (Yadav et al., 2010). Moreover, 4-fluoropiperidine has been identified as a competent binding element in the pharmaceutical field, particularly in the development of cathepsin S inhibitors (Wiener et al., 2010).

Radiotracer Development

In the field of radiotracer development, 4-fluoropiperidines have shown potential. For instance, their application in positron emission tomography (PET) has been explored, although the results were not fully satisfactory for certain specific targets (Koudih et al., 2012).

Medicinal Chemistry

4-Fluoropiperidines play a significant role as building blocks in medicinal chemistry. They have been used in the synthesis of bifunctional building blocks for fluorinated pharmaceutical compounds, offering a pathway to develop new drugs (Verniest et al., 2010). Additionally, the synthesis of cis-3-fluoropiperidin-4-ol, a variant of 4-fluoropiperidine, has been reported as an important component in medicinal chemistry (Shaw et al., 2013).
